摘要
目的 采用经超低温 (-196°)冷冻保存的同种异体动脉对大鼠行动脉移植 ,探索其形态学及免疫学方面的变化。方法 取Wistar大鼠股动脉 ,经超低温冷冻保存后行血管细胞培养及动脉移植 ;分超低温冷冻动脉移植组和自体动脉移植组。于移植后 1、2、4、8、12周 5个时间点观察动脉通畅率 ,并取材作动脉细胞存活率及组织学检测。结果 经超低温冷冻保存的动脉细胞存活率为 92 % ,比正常下降3 %。血管通畅率两组无明显差异。超低温冷冻保存的动脉复苏后 ,其外膜层正常 ,平滑肌细胞保存完整 ,细胞器正常。结论 超低温冷冻保存能降低同种异体动脉的抗原性 。
Objective To study the morphologic and immunologic changes after homologous transplantation of the cryopreserved (- 196°) artery in rats. Methods Femoral artery was harvested from Wistar rats. The cell culture of the vessel and artery transplantation were carried out after cryopreservation. The rats were divided into cryopreserved artery transplantation group and autologous one. The patency rate , the survival rate of artery cells and histological examination were observed on weeks 1,2,4,8,12 after transplantation respectively. Results The survival rate of the blood cells after cryopreservation was 92%, reducing 3% compared with the normal. No difference was found in vessel patency rate between 2 groups. when revivification of the artery was done after its cryopreservarion, there was normal adventitia and intact smooth muscle cells. Conclusions Cryopreservation may reduce the homologous artery antigenecity. Even if without the administration of the immunosuppressant agent, it is still feasible to transplant the homologous artery in rats.
