摘要
探讨影响肝癌周围肥大细胞数量的相关因素及其意义。采用二乙基亚硝胺诱发大鼠肝癌模型 ,应用光镜 ,免疫组织化学 ,核酸原位分子杂交和细胞图像分析技术 ,检测肝癌细胞转化生长因子 β(TGFβ) ,血管内皮生长因子 (VEGF) ,c- fosm RNA的表达与癌周肥大细胞 (MC)数量变化的相关性。结果显示肝癌细胞胞浆中 TGFβ,VEGF和 c- fos m RNA呈阳性表达 ,其中 TGFβ,c- fos m RNA表达的强度与癌周 MC数量变化呈平行关系 ,而 VEGF表达的强度与癌周 MC数量变化无关。结果表明 :肝癌细胞通过释放 TGFβ并通过激活原癌基因 (c- fos)以旁分泌方式或癌基因产物影响癌周
In order to investigate the correlative factors affecting the amount of mast cells surrounding hepatocarcinoma and their significance in hepatocarcinogenesis, animal models were induced by diethylnitrosamine(DENA) in rats. A series of methods such as light microscopy, immunohistochemistry, in situ hybridization, and cell image analysis were used to detect the expression of TGFβ, VEGF and c fos mRNA in hepatocarcinoma cells and their correlation with the number of mast cells surrounding carcinoma nodules. The results showed that there was a parallel relationship between the intensity of both TGFβ and c fos mRNA in hepatocarcinoma cells and the amount of mast cells surrounding the tumor nodules ( P <0 05), but VEGF expression was not related to the number of mast cells. The results suggest that the release of cytokine TGFβ by the carcinoma cells and the activation of protooncogene c fos in the carcinoma cells might affect the amount of mast cells by paracrine or protooncogene production.
