摘要
应用鸡胚绒毛尿囊膜模型 (chick embryo chorioallantoic mem brane,CAM) ,观察人骨肉瘤 OS- 732细胞系诱导血管生成过程及血管生长相关因子的表达。结果显示 ,本细胞系具有较强的促血管生成能力并表达血管内皮生长因子 (vacularendothelial growth factor,VEGF) ,碱性成纤维细胞生长因子 (basic fibroblast growth factor,b FGF) ,鸡胚绒毛尿囊膜 OS-732细胞系接种瘤细胞中血管内皮生长因子 (VEGF) ,转化生长因子β1 (Transforming growth factor,TGF-β1 )均呈阳性表达 ,而且 VEGF呈持续高表达。结果表明 VEGF、 b FGF、 TGF-β1 可能共同参与骨肉瘤 OS- 732细胞系诱导的血管生成 ,而VEGF可能起着主要作用 ,提示阻断 VEGF的作用可能影响骨肉瘤 OS- 732细胞系诱导的血管生成 ,此研究为以
The angiogenesis induced by osteosarcoma OS 732 cell line was observed by a stero microscope and light microscope in a tumor model of chick embryo chorioallantoic (CAM). The expression of angiogenic factors vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) and transforming growth factor β 1 (TGF β 1) in this cell line and xenografts was detected by immunohistochemical technique. The results showed that this cell line was strongly angiogenic, which probably is associated with production of many factors. The expression of VEGF and bFGF was positive in OS 732 tumor cell line and in xenografts on CAM. Meanwhile, the expression of TGF β 1 was negative in tumor cell line but positive in xenografts. Among those angiogenic factors, VEGF was expressed at high levels constantly. This result indicates that VEGF, bFGF and TGF β 1 are all involved in the angiogenic process of OS 732, moreover, VEGF may play a critical role among them. These findings may provide an experimental evidence for the clinical application of VEGF as a molecular target in antiangiogenic therapy of osteosarcoma.
