摘要
采用原位杂交技术结合图象分析检测常氧 (PO2 ± 2 1.3k Pa)及慢性缺氧 (P0 2 5 .3± 0 .7k Pa)培养的猪肺动脉内皮细胞 PDGF- B m RNA的表达及其对急性缺氧刺激的反应。结果 :常氧及慢性缺氧培养的肺动脉内皮细胞 (PAEC)在急性缺氧后 PDGF- B m RNA表达均增加 (P<0 .0 5 ) ,以第 4、 6代慢性缺氧组升高的幅度更大。结果表明 :慢性缺氧可增强 PAEC在急性缺氧时 PDGF- B m RNA的表达 。
In situ hybridization and image analysis system were used to investigate the expression of PDGF B mRNA in porcine pulmonary artery endothelial cells (PAEC) cultured under normoxia and chronic hypoxia conditions. The different responses to acute hypoxia were evaluated. Results: Acute hypoxia enhanced the expression of PDGF B mRNA in both the normoxic and chronic hypoxic groups. the increase being more obvious in the latter group. Conclusion: Chronic hypoxia can increase the expression of PDGF B mRNA in response to acute hypoxia in PAEC, which may contribute to the remodeling of pulmonary arteries and the development of pulmonary hypertension.
