摘要
在人泡沫逆转录病毒 (humanfoamyvirus ,HFV)原病毒全长克隆pHRSV13的基础上 ,缺失病毒基因组的env基因和bel基因 ,构建了一个表达标记基因neo和报道基因 gfp的重组人泡沫病毒载体质粒pGPSNI GFP。在与辅助质粒 p△GP共转染情况下 ,得到复制缺陷型的重组病毒颗粒GPSNI GFP。该病毒颗粒可以感染多种宿主细胞 。
A replication-defective vector (pGPSNI-GFP) base on an infections molecular clone of HFV(pHSRV13) was constructed by deletion and replacement of the env and bel genes with exprssion cassetts for neomycin-resistance and EGFP.Vector stocks were prodaced by cationic lipid reagents cotransfection of BHK-21 cells with pGPSNI-GFP and helper plasmid p △ GP.These vectors are able to transfer foreign genes into several vertebrate cells and steadily express them.
