摘要
目的 研究北京地区汉族人群中胰岛素受体底物 - 2 (IRS- 2 )基因密码子 10 5 7g/a多态性与 2型糖尿病及其中间表型的相关性。 方法 选取北京地区的中国汉族患者 110例 ,对照组 80例。用聚合酶链反应 -限制性内切酶片段长度多态性 (PCR- RFL P)的方法检测 IRS- 2基因密码子10 5 7g/a多态性。 结果 (1) IRS- 2基因密码子 10 5 7g/a多态性的 a等位基因频率在糖尿病组和对照组中分别为 2 6 .4%和 36 .3%。 (2 )糖尿病组 aa基因型频率明显低于对照组 ,分别为 5 .5 %和 18.7% (P=0 .0 16 )。L ogistic相关分析表明 :aa基因型为 2型糖尿病的保护性因素 ,OR值为 0 .2 8(95 %CI=0 .0 9~ 0 .91,P=0 .0 3)。 (3)不同基因型组间血压、血脂、胰岛素抵抗指数及胰岛 β细胞功能指数等均差异无显著性。 结论 中国北方地区汉族人群中 IRS- 2基因密码子 10 5 7g/a多态性的 aa基因型在 2型糖尿病中明显减少 ,但 2型糖尿病各种中间表型特征在不同基因型间均无明显差异。上述结果有待于进一步扩大样本量来加以确认。
Objective To study the association of insulin receptor substrate 2 (IRS 2) gene codon 1057 g/a polymorphism with type 2 diabetes and its intermediate phenotypes in Chinese Han population in Beijing. Methods The IRS 2 gene codon 1057 g/a polymorphism was detected by polymerase chain reaction and appropriate restriction enzyme (PCR RFLP) in 110 type 2 diabetic cases and 80 normal controls. Results (1) The frequency of allele a was 26.4% in case group and 36.3% in control group. (2) The aa genotype frequency of the polymorphism was significantly lower in the case group than in the control group, 5.5% and 18.7% respectively ( P= 0.016). Logistic regression analysis showed the aa genotype might be a preventive factor of type 2 diabetes, with OR=0.28 (95% CI=0.09 0.91, P= 0.03). (3) No matter in the case group or in the control group, there was no significant difference in blood pressure, blood lipid profile, islet β cell function index or insulin resistant index, etc. among three groups with different genotypes. Conclusion The aa genotype of IRS 2 gene codon 1057 g/a polymorphism has a significantly lower distribution in type 2 diabetes group than that in the control group in northern chinese population, but is not associated with any intermediate phenotypes of type 2 diabetes. The results have to be confirmed by the future study in which the volume of research sample should be enlarged.
