活血化瘀注射液对急性重型胰腺炎大鼠肺CD11b/CD18表达的影响

Effect of Huoxue Huayu Injection on the Expression of CD11b /CD18in Lung Tissue of Rats with Severe Acute Pancreatitis

目的:研究活血化瘀注射液(HHI-I)对重型急性胰腺炎(SAP)大鼠肺CD11b/CD18表达的影响。方法:将大鼠随机分为4组:假手术组;模型组;治疗组,引发SAP后即刻经颈静脉输入HHI-I,0.1mL/100g体质量,共3h;已知药对照组输入复方丹参注射液(DSHIco),剂量、时间同组3。组2、3、4分别于治疗后1、3、6及24h处死动物10只,取肺及胰腺组织标本,免疫组化法测定CD11b/CD18表达。结果:各组胰腺组织中均未检出CD11b/CD18活性。组2动物于SAP后3h见肺组织CD11b/CD18表达增加。组3动物于HHI-I治疗后1h见肺CD11b明显下降(与组2、4同时间相比p<0.05),3、6、12小时进一步降低(p<0.01);肺CD18于治疗后各阶段均明显下降(与组2、4相应时间比较p<0.01)。组4动物肺CD11b/CD18均无明显降低。结论:HHI-I可使SAP大鼠升高的肺CD11b/CD18表达明显降低;DSHIco无类似作用。

Atract:Objective:To investigate the effect of Huoxue Huayu Injection(HHI-I )on the expression of CD11b /CD18in lung tissue of rats with severe actue pancreatitis(SAP).Methods :Wistar rats were randomly divided into4groups,each containing10,50,50and50rats.1.Sham operation group.2.Model group.3.Treatment group.Intravenously HHI-I,ml per hour for 3hours,wa s administered immediately after inducing SAP through jugular vein.4.Known effective medicime group.Instead of HHI-I Dan Shen Injection composite(DSHIco)was given at the same dosage as HHI-I.10rats in group2,3,4were sacrificed at1,3,6,and24hour after treatment separately and pancreas and lung tissue samples were harvested for CD11b /CD18examination using immunochemistry technique.Results:We could not detect any activities of CD11b /CD18in pancreas.In group2,the expression of CD11b /CD18in lung of rats began to increase at3hour after SAP.For rats in group3,the activity of CD11b of lung markedly decreased at1hour after SAP(compared with group2,4P<0.05).Further decreasing was found at3,6,12hours(P<0.01).Meanwhile,the CD18expressions of lung in every time points were all decreased(compared with group2,4P<0.01).I n group4,similar changes of the expression of CD11b /CD18of lung were not noticde.Conclusion:HHI-I can significantly decline the increased expression of CD11b /CD18of lung in SAP rats.DSHIco hasn't similar effect.
