摘要
目的 :观察实验性抑郁症大鼠脑内神经生长因子含量及其 m RNA表达的变化。 方法 :采用长期不可预见性中等强度应激造成大鼠抑郁症模型。运用 openfield法观察大鼠行为学变化 ,运用免疫组化和原位杂交法观察神经元 NGF含量及其m RNA表达的变化。 结果 :与对照组相比 ,抑郁组大鼠海马和顶叶皮质神经元 NGF免疫阳性颗粒数目减少 ,着色浅淡 ;NGFm RNA杂交阳性信号减少。 结论 :实验性抑郁症大鼠海马和顶叶皮质神经元 NGF含量下降 ,NGF m RNA表达水平降低。
Objective: To observe the changes of NGF level and NGF mRNA expression in experimental depressive model of rats. Methods: The depression model of rats was established by chronic unpredictable mild stress. Openfield test was performed to detect the behavior of rats. Immunohistochemistry and in situ hybridization was used to observe the changes of NGF level and NGF mRNA expression. Results: Compared to the control group, the NGF like immunoreactivity and signals of NGF mRNA in situ hybridization of depressive group were significantly decreased. Conclusion: The levels of NGF and NGF mRNA decrease significantly in experimental depressive model of rats.
