摘要
目的 :研究缺血预处理对大鼠肝移植供肝的保护作用。方法 :将大鼠随机分为对照组 (A组 ,未行肝移植术 )、肝移植组 (B组 )和缺血预处理肝移植组 (C组 ) ,术后检测各组血清谷丙转氨酶 (AL T)、谷草转氨酶 (AST)、乳酸脱氢酶 (L DH)活性和肝组织 ATP、超氧化物歧化酶 (SOD)、丙二醛 (MDA)含量。通过光镜和透射电镜分别观察各组肝脏组织细胞形态学改变。结果 :术后 B组血清 AL T、AST、L DH活性及供肝组织 MDA含量显著高于 A组 ,而 C组升高不明显 ,并且随着缺血预处理次数增加而递减 ;B组供肝组织中 ATP和 SOD含量显著低于 A组 ,而 C组降低不明显 ,并且随着缺血预处理次数增加而接近A组含量。细胞形态学检查结果表明 ,与 B组比较 ,C组供肝组织细胞结构改变较小。结论 :适当的缺血预处理可以减轻大鼠供肝缺血再灌注损伤 ,增强对供肝的细胞保护作用。
Objective:To study the protective effects of ischemic preconditioning on donated liver during liver transplantation in rats. Methods:Rats were divided randomly into control (group A, without operation of liver transplantation), liver transplantation (group B) and ischemic precondition (group C) groups. The activity of AST, ALT, LDH and the content of ATP, MDA, SOD were all assayed after operations. The change of cell morphology were also observed under electronic and photic microscopes in all groups. Results:ALT, AST and LDH in group B were more active than in group A, while those of group C were not conspicuously active but became less active with the shortening of ischemic preconditioning time. The MDA content in group B changed in a similar way, whereas the ATP and SOD content changed oppositely, indicating ATP and SOD contents in group B was obviously lowered than that in group A. However, the decrease was not obvious in group C, which approached the same content as in group A when adding more cycles of ischemic precondition. Cell morphology changed slightly compared with group B. Conclusion:Proper ischemic precondition can relief IR injury of the donated liver, enhancing the protective impact on the cells of donated liver.
