前列腺癌错配修复基因蛋白MLH1缺失的研究 被引量:3

摘要 目的 探讨错配修复基因 (MMR)蛋白MLH1在前列腺癌细胞株及前列腺癌标本的表达及其意义。方法 利用Western印迹法检测了前列腺癌细胞株中错配修复基因蛋白的丢失情况 ;用免疫组织化学法检测前列腺癌标本中错配修复蛋白的表达。结果 在所研究的 8个前列腺癌细胞株中均发现有错配修复基因的缺失 ;前列腺癌标本中也存在错配修复蛋白MLH1表达的减少或消失。结论 前列腺癌中存在错配修复基因的缺失 ,错配修复基因的缺失直接参与了前列腺癌的发生与发展。 Objective To explore the expression and role of mismatch repair(MMR)protein MLH1 in prostate cell-lines and human prostate cancer specimens.Methods Using Western bolt analysed,we detected the loss of MLH1 in prostate cell-lines.We also immunohistochemically analysed the expression of MLH1 in human prostate cancer specimens.Results We found the loss of MMR in all prostate cell-lines and the deduction or absence of MLH1's expressions in human prostate cancer specimens.Conclusions There are defects in human prostate cancer,which influence the origination and development of prostate cancer.
