摘要
以壳聚糖微球为载体 ,经环氧氯丙烷活化后固定Anti HBsAg单克隆抗体。活化条件是 :环氧氯丙烷用量 0 3mL/g载体、活化温度 4 5℃、活化时间 4h、c(NaOH) =2mol/L ;固定化温度 3℃、固定化时间 2 4h ,活化壳聚糖与单克隆抗体的偶联率为 17.8%。患者血清体外吸附实验结果表明 ,该吸附剂可吸附除去 30 %~ 5 0 %致病抗体 。
In order to remove HBsAg(hepatitis B surface antigen) from the blood of patients with hepatitis B, the immunoadsorbent was prepared by coupling anti HBsAg monoclonal antibody to epichlorohydrin activated chitosan resins.The methods and conditions for activation of carrier,anti HBsAg monoclonal antibody coupling and adsorption with patient′s plasma were investigated.The adsorption tests showed that the immunoadsorbent could remove 30%-50% of HBsAg from the plasma when coupling rate of epichlorohydrin-activated chitosan resins to anti HBsAg monoclonal antibody was 17 8%.
