摘要
目的检测初诊儿童急性白血病bcl-2蛋白、p16蛋白和PCNA的表达,探讨细胞凋亡和增殖在白血病发生中的作用。方法运用免疫组织化学技术检测bcl-2、p16、PCNA在21例急性白血病患儿骨髓活检组织石蜡包埋切片上的表达,并与10例非白血病患儿表达情况作一对照。结果实验组bcl-2蛋白表达较对照组显著增高;实验组p16蛋白表达较对照组明显降低;实验组PCNA表达较对照组显著增高。结论bcl-2抑凋亡作用增强和p16抑增殖作用减弱在儿童白血病发生发展中具有重要作用,PCNA则可作为细胞增殖状态的标志。
Objective To study the expression of bcl 2?p16 and PCNA in newly diagnosed childhood acute leukemia and to illustrate the role of apoptosis and proliferation in the pathogenesis of leukemia. Methods The expression of bcl 2?p16 and PCNA in bone marrow were studied by immunohistochemistry in21 cases of children with newly diagnosed acute leukemia selected as test groupand in 10 cases of children without leukemia served as control group. Resultes he expression rates of bcl 2 protein in test group was significantly higher than control group; The expression rates of p16 protein in test group was significantly lower than control group; The percentage of PCNA positive cells in test group was significantly higher than control group. Conclusion Both apoptosis weakened by bcl 2 protein high expression and proliferation strengthened by p16 protein deletion may play an important role in the pathogenesis of childhood acute leukemia, and PCNA may reflect the activity of cells proliferation.
