表皮生长因子对急性胰腺炎大鼠的保护作用

摘要 目的探讨表皮生长因子(EGF)对急性胰腺炎(AP)大鼠胰腺炎相关蛋白(PAP)基因表达的影响及对损伤胰腺的保护作用。方法雄性SD大鼠24只,经胰胆管逆行注入3.5%牛磺胆酸钠(1.0ml/kg),诱导AP模型,然后随机分为3组:对照组,AP组及EGF组,每组8只。24小时后,检测血清淀粉酶,观察胰腺组织病理学变化,采用原位杂交组织化学技术结合计算机图象分析法,测定胰腺细胞PAPmRNA水平。结果EGF组动物胰腺组织病理学损害明显较AP组轻微,血清淀粉酶值显著低于AP组(P<0.01),胰腺PAP基因表达阳性面积率显著低于AP组(P<0.01)。结论EGF可以减少AP大鼠PAP基因表达阳性面积率,对损伤胰腺具有保护作用。 Objective To evaluate the effects of epidermal growth factor(EGF)on pancreatitis-associated protein(PAP)gene expression and on injured pancreas in rats with acute pancreatitis.Methods Twenty-four male Sprague-Dawley rats that underwent injection of3.5%sodium taurocholate solution into the pancreatic duct were randomly divided into control group,AP group and EGF group.There were8rats in each group.Rats were sacrificed on24h after induced pancreatitis.Serum amylase and pancreatic pathology were determined,PAP-mRNA level was measured with in situ hybridization histochemistry and image processing and analysis system.Results In EGF group,pathological injury in pancreas was significantly milder than that in AP group;Serum amylase was significantly lower than that in AP group(P<0.01);PAP-mRNA level decreased significantly than that in AP group(P<0.01).Conclusions EGF decreased PAP-mRNA gene expression and might protect pancreas against injury in rats with acute pancreatitis.
