内皮抑素抑制人脐静脉内皮细胞分泌IL-6和IL-8的实验研究

摘要 目的 :研究内皮抑素对体外培养的人脐静脉内皮细胞 (HUVEC)分泌白细胞介素 6 (IL 6 )和白细胞介素 8(IL 8)的抑制作用。 方法 :在建立HUVEC体外培养模型的基础上 ,将细胞分为自然分泌组和内皮抑素组进行培养 ,收集 1~ 9天的培养上清液 ,离心。采用双抗体夹心ELISA法对上清液中IL 6和IL 8进行定量测定。 结果 :在自然分泌组HUVEC培养上清液中检测出IL 6和IL 8,证实了在自然状态下内皮细胞能产生一定量的IL 6和IL 8,而且随着内皮细胞培养天数的增加 ,IL 6和IL 8的分泌量也随之增加。当细胞进入对数增长期后 ,IL 6和IL 8的分泌量接近峰值 ,分别为 (2 979.32± 19 6 5 )pg/ml和 (6 0 18.87± 5 6 .74)pg/ml。内皮抑素组较自然分泌组HUVEC培养上清液中的IL 6和IL 8水平存在显著差异。 结论 :内皮抑素能够很好地抑制内皮细胞的生长和增殖 ,降低内皮细胞的生物活性。 Objectives:To investigate the inhibitory effects of endostatin on IL 6 and IL 8 release from human umbilical vein endostelial cells(HUVEC). Methods: HUVEC culture model was founded. The human endothelial culture supernatant(HECS) was harvested from primary culture medium for 1~9 days and was centrifuged. The cells were divided into two groups: normal control and endostatin. IL 6 and IL 8 in HECS was measured using sandwich enzyme linked immunosorbent assay(ELISA)method. Results: IL 6 and IL 8 were detected in HECS without stimulating factors in the endostelial culture. The amounts of IL 6 and IL 8 increased along with the cell culture time. At 6th day the amount reached their peaks (2 979.32±19.65)pg/ml and (6 018.87± 56.74)pg/ml. In the endostatin group, the amounts of IL 6 and IL 8 were decreased. Conclusions: Endostatin could inhibit the growth, reproduction and the biological activity of endothelial cells.
