Immunoblotting Study of Thyroid Autoantibodies in Graves' Disease

目的 :研究Graves’病 (Graves’disease ,GD)甲状腺自身抗体的异质性。方法 :以正常、GD及桥本氏甲状腺炎患者 (HT)甲状腺组织作为抗原 ,运用Westernblot检测 2 0例GD甲亢期及 2 0例GD缓解期患者的甲状腺自身抗体 ,并以正常阴性血清及标准阳性血清 [甲状球蛋白 (TGAb) +、甲状腺过氧化物酶抗体 (TPOAb) +、TSH受体抗体 (TRAb) +]作为对照。结果 :GD血清可与多种甲状腺抗原反应 ,每个患者出现的阳性条带带型均不一样。GD血清在HT及GD甲状腺组织抗原泳道出现的阳性条带数明显多于正常甲状腺组织抗原泳道出现的阳性条带数 (P <0 .0 1) ,且有的条带不出现于正常甲状腺组织抗原泳道。与标准阳性血清出现的阳性条带相比 ,GD血清出现更多的条带。结论 :GD甲状腺自身抗体存在异质性 ,GD时存在除TGAb、TPOAb、TRAb以外其它的甲状腺抗体 ,甲状腺细胞中某些蛋白的变异可能是导致GD时产生甲状腺自身抗原的原因之一。

Objective:To study the heterogeneity of thyroid autoantibodies in Graves' disease. Methods:Using thyroid tissues of the normal, and patients with Graves' disease and Hashimoto's thyroiditis as antigen,we detected the thyroid antibodies by Western blot in 20 patients with Graves' disease in hyperthyroidism and 20 patients with Graves' disease in remission.The sera of normal negative and standard positive (TGAb+, TPOAb+, TRAb+) were used as the control. Results:The sera of patients with Graves' disease can react with many thyroid antigens in Western blot.The types of positive bands were different in each patient with Graves' disease.The number of positive bands appearing in electrophorestic lane of tissues of Graves' disease and Hashimoto's thyroiditis was much larser than that in normal thyroid.Some positive bands didn't appear in electrophorestic lane of tissues of normal thyroid. Compared with the sera of normal negative and standard positive, GD sera appear to be more positive bands. Conclusions:In addition to TGAb,TPOAb and TRAb,there were new thyroid antibodies in Graves' disease and there was heterogeneity in thyroid autoantibodies. Moreover, the variation of some proteins in thyroid cell is possibly one of the causes of appearance of thyroid autoantigen in Graves' disease.
