摘要
本研究从湘西野生蓖麻籽中提取了两种植物毒蛋白ricin Ⅰ、ricin Ⅱ,其分子量分别为65、67KD,LD_(50)分别为0.20、0.24ug/小鼠。用本室制备的抗大肠癌单克隆抗体Hb_3作为导向载体,与蓖麻毒蛋白A肽链交联,制备杂交分子Hb_3—RTA。SDS—PAGE分析表明,每个Hb_3上平均结合4.9个RTA。初步细胞毒试验显示,交联物Hb_3—RTA对大肠癌细胞HRT-18具有较强杀伤作用,而对正常人淋巴细胞杀伤作用较小。
Two plant toxic proteins, ricin Ⅰ and ricin Ⅱ, were isolated and purified from Xiangxi castor beans.The molecular weight of ricin Ⅰ, Ⅱ are 65, 67 KD respectively.By covalent coupling the A chain (RTA) from ricin Ⅰ to a monoclonal antibody Hb3, an immunotoxin Hb3-RTA was obtained.The conjugation Hb3-RTA showed the structure of native toxin, every Hb3molecule was linked on average 4.9 RTA. It showed cytotoxic activity to colorectal carcinoma cell HRT-18 in culture, but it was not toxic for human lymphocyte.
