产紫杉烷类物质的链格孢1011菌种的培养和发酵条件 被引量:4

摘要 用真菌学方法对产紫杉烷类物质的链格孢1011菌种培养和发酵条件进行了研究,结果表明:该菌易培养,生长发育迅速。用固体培养基培养,划破菌落或变温交替培养,可促进大量产孢。发酵条件优化结果:种子培养的最佳时间为 3d,发酵最佳培养时间为 8d,前体物苯甲酸纳可促进化合物Ⅰ的合成,通过氮、碳源优化组合,发酵培养基的最佳氮、碳源每100mL萄糖2.2g,花生饼粉2.6g,黄豆饼粉0.2g,淀粉2.2g,蛋白胨0.48g,酵母膏0.5g,氯化铵0.58g,硫酸镁0.3g,磷酸二氢钾0.3g。采用最佳发酵条件优化培养,可使发酵产物化合物Ⅰ的组分比原始培养条件下产量提高了262%。 The conditions of slope culture and liquid fermentation of taxoids - produced endophytic fungi Alternaria sp. 1011 were studied through mycological methods. The results prove that the strain grows fast and is easily cultivated. Lacerating the-colony or shifting the culture temperature during the solid slope culture can promote it to produce more spores. The opti-mum culture conditions are that the optimum culture time of seed and fermentation are 3 days and 8 days respectively and as a precursor, benzoate can promote the biosynthesis of compound Ⅰ. The optimum fermentation media obtained through C、N source uniform test are: glucose 2. 2 g, soluble starch 2. 2 g, peanut meal 2. 6 g, soya meal 0. 2 g, peptone 0. 48 g, yeast extract 0. 5 g, ammonium chloride 0. 58 g, magnesium sulfate 0. 3 g, potassium dihydrogen phosphorate 0. 3 g, water 100 mL, pH natural, sterilization 121℃ × 30 min. Under the optimized culture condition, the compound Ⅰ yield increases by 262% compared to the control.
