人精子细胞膜透明质酸酶对肾上腺癌细胞SW13增殖影响的研究 被引量:2

摘要 目的 探讨人精子细胞膜结合型透明质酸酶 (PH2 0 )对人肾上腺癌细胞SW 13生长的影响。方法将人全长PH2 0cDNA转染到体外培养人肾上腺癌细胞SW 13细胞株中 ,将细胞置入软琼脂培养板中 ,研究集落形成的改变 ;再将同样的细胞种植到 10d龄鸡胚绒毛尿囊膜 (CAM )上 ,研究肿瘤生长大小的变化 ;细胞培养在2 4孔培养板中 ,采用细胞计数方法研究细胞的生长曲线。结果 转PH2 0基因的SW 13细胞在软琼脂培养板中的集落形成能力明显增强 ,CAM上的SW 13增殖程度明显高于对照组 ,转PH2 0基因后SW 13细胞生长曲线上升。结论 PH2 0可促进人肾上腺癌细胞SW 13的增殖 ,提高SW Objective To study the effects of human spermatic membrane bound hyaluronidase (PH20) on SW13 (human adrenal carcinoma) cells growth. Methods Full length cDNA of human PH20 was transfected into SW13 cell line, and the transfectants were therefore implanted onto chorioallantoic membrane (CAM) and cultured in soft agar plates. Also, cell proliferation curve was studied after the cell was transfected with PH20 cDNA by counting the cell numbers on 24 well culture plates. The following was done for the tumor growth study: (1) weighing and measuring the tumors from CAM; (2) counting the colonies on soft agar larger than 60 μm ;(3) counting cell numbers on 24 wells plates every day. Results Comparing with the mock transfectants, the exografts of SW13 PH20 were larger than SW13 transfected with empty vectors (pcDNA3). Colonies were much increased in SW13 PH20 group than in the SW13 pcDNA3 control group, paralleled with a same increased level of cell proliferation curve. Conclusion The results suggest that membrane bound hyaluronidase (PH20) may play an important role in promoting human tumor growth.
