摘要
目的 :探讨釉原蛋白的作用机制 ,为临床应用釉原蛋白进行龋病的治疗奠定基础。方法 :采用细胞培养、放射免疫测定和碱性磷酸酶 (AKP)检测等实验技术 ,探讨釉原蛋白Am对牙髓外胚间充质细胞的作用。结果 :通过观察Am纯化蛋白可溶性上清液对体外培养的牙髓细胞的骨钙素含量和碱性磷酸酶活性的影响 ,发现Am可增加骨钙素含量 ,增强碱性磷酸酶活性 ,并呈剂量依赖性 ,在 10 0ng/ml浓度下两者分别增加 3和 4倍。结论
AIM: To evaluate the function of amelogenin protein (Am) on development of dental enamel in order to use Am to treat dental caries reasonably. METHODS: Cell culture, radioimmunoassay and alkaline phosphatase detection were used to research the effect of Am to human dental pulp cells on osteocalin synthesis and alkaline phosphatase activation. RESULTS: Examination of Am to human dental pulp cells in vitro showed that Am was able to stimulate the induction of alkaline phosphatase and osteocalin synthesis in a dose-dependent manner. The effective dose range for Am in the assays performed in our study was 1× 10 2~1× 10 3 ng/ml. CONCLUSION: Am might be effective in stimulating human dental pulp cells to differentiation and mineralizaton.
