hPER1-NLS: study of a novel penetrating peptide

摘要 目的 研究hPER1-NLS多肽的穿膜特性。方法 用固相法化学合成hPER1-NLS多肽及对照多肽 ,用免疫组化方法观察hPER1-NLS的穿膜能力及在细胞中的定位 ,并对其穿膜的特性做初步研究。结果 hPER1-NLS对细胞膜有穿透作用 ,主要定位在细胞核中。且在不同作用环境条件下 ,都具有穿膜能力。结论 hPER1-NLS也是一种MPP 。 Objective To study property of penetrating membrane of hPER1-NLS. Method hPER1-NLS and other peptides were synthesized by solide-phase method and immunohistochemical technique was used to confirm its targeting of intracellular or intranuclear. Property of penetrating membrane of hPER1-NLS was studied in different conditions. Results hPER1-NLS possessed penetrating ability in different conditions and demonstrated intense focal staining in the nuclear plasma as well as in the nucleus. Conclusion It is implied that hPER1-NLS is also MPP, suggesting that hPER1-NLS is used in gene therapy and study of the mechanism of penetrating membrane.
