摘要 目的 研究 p2 1CIP1 /WAF1 及 PCNA在人前列腺癌标本中的表达及相关性 ,阐述它们与前列腺癌病理分级及临床分期的关系 .方法 以 36例确诊前列腺癌石蜡包埋存档标本作为研究对象 ,采用免疫组化 SABC法对其 p2 1CIP1 /WAF 1及PCNA进行检测 ,并应用图像分析仪判定 ,统计学处理 ,对前列腺癌的病理分级及临床分期进行了对比分析及相关性研究 .结果  p2 1CIP1 /WAF 1 及 PCNA的免疫组化阳性染色为棕黄色和 /或棕褐色 ,定位于细胞质或细胞核 .所得数据均经统计学处理 .在不同病理分级及临床分期之间差异均有显著性 ,同时还发现 PCNA与 p2 1CIP1 /WAF 1存在显著负相关性 .结论 p2 1CIP1 /WAF1表达与前列腺癌组织的病理分级及临床分期呈负相关性 ,在发病机制中可能涉及到 p2 1CIP1 /WAF1 和 PCNA调节通路的异常 .同时 ,作为一种检测方法 。 AIM To research the expression and correlationof p21 CIP1/WAF1 and PCNA in human prostate cancer, for the purpose of illustrating their relations to the pathological grading and clinical phases. METHODS Thirty six subjects were collected, which were the paraffin embedding samples that had been diagnosed as prostate cancer. p21 CIP1/WAF1 and PCNA was tested by immunohistochemical SABC method. By using image pattern analyzer and through statistical methods, we did the comparative analysis and correlation research for the pathological grading and clinical phases of the prostate cancer. RESULTS The immunohistochemical positive staining of p21 CIP1/WAF1 and PCNA was brown or dark brown, which were located in cytoplasm and nucleus. All data were treated through statistic analysis. As a result, the significant differences were found in different pathological grading and clinical phases. Meanwhile, we also found there was negative correlation between PCNA and p21 CIP1/WAF1 . CONCLUSION The above mentioned research shows that p21 CIP1/WAF1 is involved in the occurrence and development of prostate cancer. As for the pathological mechanism, it is related to the abnormal of p21 CIP1/WAF1 and PCNA regulating path. Therefore, as a testing method, the research can be an index for estimating the level of malignant and progression of the prostate cancer, and expectedly, provide some references for diagnosis and treatments.
