The expression of RhoCmRNA and RhoC protein in human gastric carcinoma and implication

摘要 目的 探讨胃癌肝转移中胃癌组织及肝转移癌组织中RhoCmRNA及蛋白的表达和意义。方法 采用逆转录 聚合酶链式反应技术 (RT PCR)分析 2 8例胃癌患者的癌组织 (GC)及癌旁胃粘膜组织 (PGMT)、肝转移癌 (ML)组织中RhoCmRNA及蛋白的表达 ;免疫组织化学染色分析RhoC蛋白的表达情况。结果 GC组织中RhoCmRNA吸光度相对值为 0 .13 5± 0 .0 65 ;PGMT的吸光度相对值为 0 .118± 0 .0 74,两者差异无显著性 (P >0 .0 5 ) ;ML中RhoCmRNA吸光度相对值为 0 .64 1± 0 .113 ,与GC相比 ,差异有显著性 (P <0 .0 5 )。RhoC蛋白的表达中 ,肝转移灶者、胃癌组织、癌旁胃粘膜组织的灰度值分别为 (12 8.5 2 3± 11.486)U ,(14 6.694± 7.0 79)U ,(162 .3 5 1±14 .612 )U ,经t检验 ,差异有显著性 (P <0 .0 5 ) ;ML中RhoCmRNA及蛋白吸光度相对值明显高于GC(P <0 .0 5 )。结论 RhoCmRNA及蛋白的表达与胃癌转移有关 ,可作为判定胃癌转移及预后的指标。 Objective To study the expression of RhoCmRNA and RhoC protein in human gastric carcinoma (GC),the paracarcinoma gasric mucosa tissues (PGMT) and metastasis of liver (ML).Methods Reverse transcription polymerase chain reaction was used to detect the expression of RhoCmRNA in the GC,PGMT and ML of 28 patients with gastric cancer.Results The optical density (A) of the expression of RhoC mRNA in GC was 0.135±0.065,not significantly different that in PGMT ( 0.118± 0.074,P>0.05).But the expression of RhoC protein in GC (146.694±7.079) U was significantly higher than in ML (128.523±11.486) U(P<0.05) and PGMT (162.351±14.612).The expression of RhoC protein in ML (162.351±14.612) was significantly higher than in GC(P< 0.05) .Conclusion The expression of RhoCmRNA in GC was correlated with metastasis and it may be a good marker for prognosis and metastasis of gastric cancer.
