肝细胞癌与乙肝病毒基因整合和p16基因表达的相关性研究 被引量:1

The relationship between the integration of hepatitis B virus gene and the expression of p16 gene and hepatocellular carcinoma

摘要 目的 探讨乙型肝炎病毒 (HBV)DNA整合与p16的关系及其在肝细胞癌发生发展中的作用。方法 聚合酶链式反应 (PCR )检测 3 5例肝癌、癌旁组织及 3 1例肝硬化组织中HBVDNA及其整合情况 ,以Southernblot证实 ;逆转录 聚合酶链式 (RT PCR)检测组织中 p16mRNA。结果 有HBVDNA整合的肝癌、癌旁及肝硬化组织中X基因整合率分别为 10 0 .0 %、81.8%及68.4% ,三者差异有显著性 (P <0 .0 5 ) ;p16mRNA的表达缺失率分别为 5 1.4%、3 1.4%、2 2 .6% ,三者差异有显著性 (P <0 .0 5 )。结论 HBVDNA整合与 p16基因表达改变有相关性 ,两者在肝癌的发生发展中发挥重要作用。 Objective To investigate the relationship between the integration of hepatitis B virus (HBV) DNA,the expression of p16 gene and the carcinogenesis and development of hepatocellular carcinoma (HCC).Methods 35 cases of HCC and 31 cases of liver cirrhosis (LC) were studied.Polymerase chain reaction (PCR) method was applied to examine HBV DNA in serum and tissues and detect HBV DNA integration confirmed by southern blot analysis.The expression of p16 gene in tissues was detected by reverse transcriptase PCR (RT PCR).Results The integration rate of HBV DNA in HCC,adjacent region and LC was 71.4 %(25/35),62.9 %(22/35),61.3 %(19/31)respectively,with the difference being not significant among them(P>0.05).But the integration rate of HBVX gene in these three kinds of tissues integrated by HBV DNA was 100.0 %(25/25),81.8 %(18/22)and 68.4%(13/19)respectively,with the difference among them being statistically significant (P<0.05).The expression rate of p16 gene in the above tissues was 51.4 %(18/35),31.4 %(11/35),22.6 %(7/31)respectively,with the difference being significant(P<0.05).Conclusion There was correlation between the expression loss of p16 gene and HBV DNA integration,which may play important roles in the carcinogenesis and development of HCC.
