摘要
目的 探讨骨形态发生蛋白 (BMP)对骨骼肌卫星细胞粘附特性的影响。方法 体外获取与培养骨骼肌卫星细胞 ,分别用 0、5 0、10 0、5 0 0、10 0 0 μg/L的BMP诱导培养基诱导培养 48h。通过荧光法测定接种后 1h的粘附细胞率 ,利用放射免疫法测定细胞层粘连蛋白的含量。结果 BMP可促进骨骼肌卫星细胞的粘附 ,在 5 0 0 μg/L的浓度时粘附率最高 ,当BMP浓度进一步增加时 ,细胞粘附率不再增加 ;BMP诱导后可促进骨骼肌卫星细胞层粘连蛋白的表达 ,当BMP浓度为5 0 0 μg/L时这种作用最强。 结论 BMP可增强骨骼肌卫星细胞的粘附特性 ,其机制与层粘连蛋白的表达增高有关。
Objective To study the effect of bone morphogenetic protein (BMP) on the adhesive characteristics of skeletal muscle satellite cells.Methods Skeletal muscle satellite cells were harvested and cultured in vitro.The 0,50,100,500,1?000?μg/L BMP were used to induce skeletal muscle satellite cells for 48?h.The attached cells was calculated by fluorescence method at 1 h after seeding.The laminin of cells was measured by radioimmuno assay(RIA).Results The number of attached cells were increased after the cells were induced with BMP with the concentration below 500?μg/L,and reached the highest number when BMP was 500?μg/L.The number of attached cells were not increased with the increase of BMP concentration.The laminin of cells was increased after the cells were induced with BMP,and reached the highest when BMP was 500?μg/L.Conclusion BMP enhances the adhesive characteristics of skeletal muscle satellite cells,which might be contributed to the increased expression of laminin.
