摘要
目的 探索日本血吸虫新的疫苗候选分子对小鼠的保护性免疫效果。方法 利用已构建的真核表达质粒 p BK-Sj14 -3 -3在大肠杆菌 BL2 1内表达的日本血吸虫 (大陆株 ) 14 -3 -3 epsilon同型融合蛋白质 ,免疫 6周龄 BAL B/c雌性小鼠 ,免疫 3次 ,每次间隔 2周 ,第 3次免疫后 2周感染尾蚴。42 d后剖杀小鼠 ,计算其减虫率和肝减卵率。结果 各免疫组与对照组相比 ,均获得了部分抗血吸虫保护性免疫力 ,第 1、2、3各免疫组的减虫率分别为 3 2 .7% (P<0 .0 5 )、3 0 .7% (P<0 .0 5 )、2 7.5 %(P<0 .0 5 ) ;其肝减卵率分别是 48.5 % (P<0 .0 5 )、43 .1% (P<0 .0 5 )、2 8.2 % (P<0 .0 5 )。结论 首次证明日本血吸虫 14 -3 -3 epsilon同型重组融合蛋白可诱导小鼠抗血吸虫感染的部分保护性免疫力。
Objective To research the effect of protective immunity for a new vaccine candidate of Schistosoma japonicum (Sj) in mice.Methods Using the constructed eukarytic expression plasmid pBK Sj14 3 3, fusion protein of 14 3 3 epsilon isoforms of Sj(Chinese strain) was expressed by E.coli BL21. Six week BALB/c female mice were immunized with the fusion protein, total three times, and two weeks after third immunity, the mice were infected with cercariae of Sj. The mice were killed after 42 days and compared with the control group. The worm reduction rates and liver egg reduction rates were calculated. Results A part protective immunity was obtained in every immune group. In first, second, third group, the worm reduction rates were 32 7%(P<0 05)?30 7%(P<0 05)?27 5%(P<0 05) respectively; the egg reduction rates 48 5%(P<0 05)?43 1%(P<0 05)?28 2%(P<0 05) respectively. Conclusion Fusion protein of Sj14 3 3 epsilon isoforms are partially effective against S. japonicum.
