原位核酸杂交检测膀胱癌中结核菌L型DNA的意义 被引量:1

摘要 目的 探讨结核菌L型DNA表达在膀胱移行细胞癌发生与发展中的意义。方法 改良抗酸 (IK)染色和免疫组化染色 (S P法 )检测 6 8例膀胱癌组织中抗酸菌L型和结核菌抗原 ;原位核酸杂交 (ISH)检测 2 0例膀胱癌细胞内结核菌L型DNA。结果 抗酸菌L型阳性率 35 .2 % ,结核菌抗体 (BCG)阳性表达率 36 .7%。 4 0 %膀胱癌细胞内有结核菌L型DNA阳性信号 ,膀胱癌Ⅰ、Ⅱ、Ⅲ级阳性表达率分别为 2 0 %、30 %、80 % (P <0 .0 5 ) ,随着膀胱癌级别的增加 ,结核菌L型感染率显著增高。改良抗酸 (IK)染色、免疫组化染色和原位核酸杂交结果具有一致性。结论 膀胱癌中常伴有结核L型感染 ,结核菌L型DNA可进入膀胱移行细胞内 ,引起细胞增生和癌变 ,提示结核菌L型感染在膀胱癌发生、发展中起重要作用。 Objective To evaluate the signficance of tubercle bacillus L?form DNA expression in pathegenesis and prognosis of bladder transitional cell carcinoma(BTCC). Methods 68 cases of BTCC were detected with Intensifed Kingoun(IK) staining, BCG immunohistochemistry (IH) and 20 cases of BTCC were examined in situ hybridization(ISH) . Results The total positive rate of antiacid bacillus L?form was 35.2% (24/68), BCG was 36.7% (25/68) in BTCC. The positive expression rate of tubercle bacillus L?form DNA was 40% in BTCC, WHO grade Ⅰ, Ⅱ, Ⅲ were 20%,30%, 80% respectively.With the increase in WHO grade, the infection rate of tubercle bacillus L?form increased (P<0.05). The results in IK, BCG IH and ISH were highly concident. Conclusion BTCC is frequently accompany by infection of tubercle bacillus L?form, tubercle bacillus L?form DNA where is in ladder transitional cell may result in proliferation and malignant transform of ladder transitional cell. It suggested that the infection oftubercle bacillus L?form may play an important role in the development and malignant progression of BTCC.
