摘要
目的:建立适合甲巯咪唑血药浓度快速测定的HPLC法。方法:色谱柱:Hypersil ODS柱(150 mm×4.6 mm,5μm);流动相:甲醇-0.05%乙酸溶液(1:99);波长:252 nm。结果:该方法在含甲巯咪唑血浆浓度0.1~2.0μg/ml及2.0~50.0μg/ml范围内定量线性良好(r分别为0.9994和0.9999),最低检测限为1ng。精密度和回收率均良好。结论:本方法适用于服用甲巯咪唑及卡比马唑的临床病人的血药浓度测定,以便进行临床药学研究。
Objective: To establish a rapid HPLC method for determining the concentration of thiamazole in human plasma. Methods: A Hypersil ODS column(150 mm × 4. 6 mm, 5 μm)was used. The mobile phase was methanol-0. 05% acetic acid (1:99). The wavelength of detector was 252 nm. Results: The method gave a good linearity of thiamazole concentrations in plasma ranging from 0. 1 to 2. 0 μg/ml(r=0. 999 4) and 2. 0 to 50. 0 μg/ml(r=0. 999 9). It also had good precision and recovery. Conclusion: This method could be used to determine thiamazole concentrations in patients receiving thiamazole or carbimazole.
