摘要
目的 研究巨噬细胞氧化低密度脂蛋白的定位。方法 以培养的小鼠巨噬细胞为模型 ,与 10 0 μg/mL低密度脂蛋白共培养 ,将其交联后分离不同膜成分 ,通过膜上髓过氧化物酶和低密度脂蛋白含量测定 ,研究巨噬细胞氧化低密度脂蛋白的细胞定位。结果 在分离得到的不同膜成分中 ,5 %蔗糖密度梯度液部分髓过氧化物酶和低密度脂蛋白含量分别比 10 %部分高出 7.6 6 7和 2 1倍。结论 巨噬细胞对低密度脂蛋白的氧化发生在细胞膜上的“Microdomain”中。
Objective To investigate the cellular orientation of LDL oxidation by macrophage. Methods Based on the model of mouse macrophage, macrophage was incubated with LDL(100 μg/mL),and cell membrane component parts were isolated after cross linking them together. To investigate the cellular orientation of LDL oxidation by macrophage, the content of myeloperoxidase and LDL on membrane were determined. Results In the before mentioned isolated membrane composition, the content of myeloperoxidase and LDL in the 5% sucrose den sity gradient solution was 7.667 which was 21 times higher than that found in the 10% sucrose density gradient solution. Conclusion The LDL oxidation by macrophage occurs on the 'Microdomain'.
