VEGF在非小细胞肺癌中表达与血管生成关系及临床意义 被引量:6

摘要 目的 采用免疫组化方法观察血管内皮生长因子 (VEGF)及血管内皮细胞膜抗原CD34在非小细胞肺癌(NSCLC)组织、肺炎性假瘤及正常肺组织中的表达状况。方法 用抗VEGF多克隆抗体及CD34单克隆抗体作免疫组化染色 ,免疫标记物阳性细胞和癌组织中微血管密度 (MVD)计数。结果  81例NSCLC组织上VEGF表达的总阳性率为 72 .84 % ,VEGF在肺癌细胞中主要分布于胞浆、胞膜 ,少量胞外基质也有阳性表达。鳞形细胞癌阳性细胞呈弥散或局灶分布 ,腺癌则呈腺泡状分布。VEGF表达强度同非小细胞肺癌的MVD值紧密相关 ,随VEGF表达强度增高 ,MVD值亦显著增高 (P <0 .0 0 1)。结论VEGF的表达和MVD与NSCLC的发生、发展、转移关系密切 。 Objectives To investigate the involvement of VEGF in the occurrence and development of non small cell carcinoma of lung by examination of the expression of VEGF and CD34 between NSCLC, inflammatory pseudotumor of lung and normal lung tissue with immunohistochemical staining, to study the relationship between VEGF expression and angiogenesis in NSCLC.Methods Immunohistochemical staining was performed by anti VEGF polyclonal antibody and anti CD34 monoclonal antibodies, and the degree of antibody reactivity with individual tissue sections was considered to be positive if unequivocal brown yellow staining of membrane or cytoplasm was seen in tumor cells. Results The total positive rate of 81 patients is 72.84%,VEGF was mainly distributed in cytoplasm and membrane of tumor cell,some tumors demonstrated positive staining in the basal lamina or extracellular stroma ,so did some normal tissue.Positive cell showed diffuse or local diffusion in SQCC and surrounded around adeno alveola in ADC.It was found that there was a close relationship between MVD and VEGF expression in NSCLC and MVD increased with the higher expression of VEGF ( P <0.001 ). Conclusion Expression of VEGF is closely related to the occurrence and development of NSCLC and plays an important role in neo angiogenesis, it may act as a diagnostics marker for clinical biological characteristic in NSCLC.
