老年慢性阻塞性肺疾病发作期患者T淋巴细胞核仁区酸性非组蛋白表达活性的分析

THE SIGNIFICANCE OF NUCLEOLAR ORGANIZING REGION ASSOCIATED PROTEINS OF T LYMPHOCYTES IN THE OLD CHRONIC OBSTRUCTIVE PULMONARY DISEASE AT ITS ACUTE PHASE

目的 通过对老年慢性阻塞性肺疾病 (COPD)发作期患者T淋巴细胞Ag -NORs酸性非组蛋白表达活性的分析 ,探讨Ag -NORs与老年COPD患者细胞免疫功能的关系 ,并进一步探讨Ag -NORs在老年COPD急性发作期中的临床应用价值。方法 利用免疫图像分析系统、细胞培养、硝酸银染色等方法 ,对 30例健康老年人及 40例老年COPD急性发作期外周血T淋巴细胞Ag -NORs的酸性非组蛋白的蛋白质进行染色分析。结果 老年COPD急性发作期患者Ag -NORs非组蛋白活性 (5 72± 0 77) % ,明显低于健康老年人 (P<0 0 5 ) ;对照组中 2 6 7%为I·S % >8 0 0 %的高活性表达 ;而老年COPD组仅为 7 5 % ,二组比较有显著性意义(P <0 0 5 ) ;I·S % <5 0 0 %的极低活性分析中 ,对照组有 4例 (13 3% ) ,老年COPD组有 19例 (4 7 5 % ) ,有非常显著性意义 (P <0 0 1)。结论 老年COPD急性发作期患者外周血T淋巴细胞Ag -NORs非组蛋白的表达活性明显低下 ,推测老年COPD患者T淋巴细胞核内核糖体的装配数量明显减少 ,核糖体合成蛋白质的数量不足 ,功能异常 ,并进一步反映细胞免疫功能低下 ,是老年COPD患者易患各种感染并且急性发作的一个主要原因。因此 ,Ag -NORs是老年COPD患者的诊断 。

Purpose To investigate the significance of nucleolar organizing regions(NORs) associated proteins of T lymphocyte in the old chronic obstructive pulmonary disease(COPD) at its acute phase.Methods Analysis of the silver staining proteins in NORs(Ag-NORs) in 40 patients with the old COPD.In contrast with the COPD group,30 healthy old people were observed as above.Result Compared with the healthy old people group,the activity of Ag-NORs were obviously decreased(P<0.05).Both 26.7% in healthy old people and 7.5% in the old COPD,above 8% I.S% values have an obvious difference(P<0.05).Meanwhile,both groups of 13.3%(4 cases)in healthy old people and 47.5%(19 cases)in the old COPD less than 5.00% I·S% values have a significant difference(P<0.01).Conclusion Ag-NORs associated proteins,in T lymphocytes were apparently decreased in the old COPD at its acute phase.It is supposed that the quantity of ribosomes was obviously decreased and lack of the associated proteins.The COPD mainly result from descent and disorder of the cellular immunity.It is available for the diagnosis,evaluation of therapeutic effect and prognosis in COPD
