摘要
目的 初步观察胚胎心肌细胞移植于大鼠急性心肌梗死周边区后第 72小时内的存活情况。方法 移植组结扎成年大鼠冠状动脉的同时在预测的梗死周边区注射培养的胚胎心肌细胞 ,对照组在正常大鼠的心肌内注射培养的胚胎心肌细胞 ,空白组结扎冠状动脉的同时在与移植组相同的部位注射等量的培养液。在移植后第 72小时采集标本 ,进行普通H -E染色及免疫组化染色 ,显微镜观察。结果 在移植组和对照组都可以看到移植的胚胎心肌细胞的存在 ,空白组未发现有移植的胚胎心肌细胞。结论 移植后
Objective To assess the capacity of fetal cardiomyocytes to survive in the border zone of acute myocardial infarction of adult rat within 72 hours.Methods In the treatment group and the sham-operated group,the left coronary artery was ligated,followed by the injection of the cultured fetal cardiomyocytes and culture medium respectively.In the control group,the fetal cardiomyocytes were injected into the normal myocardium.At seventy-two hours after cell transplantation,the hearts were harvested.The heart slices were processed for histological(H-E) and immunohistological(anti-BrdU) techniques and photographed.Results Cultured fetal cardiomyocytes could be found both in the normal myocardium of control animals and in the border zone of acute myocardial infarction after transplantation.Whereas,there′s no fetal cardiomyocyte present in sham-operated animals.Conclusion Cultured fetal cardiomyocytes can survive in normal myocardium and in the border zone of acute myocardial infarction within seventy-two hours after transplantation.
