UTI和MPG在鼠肝脏缺血再灌注损伤保护效应的实验研究 被引量:1

摘要 目的 :研究蛋白酶抑制剂 -乌司他丁 (urinarytrypsininhibitor ,UTI)和氧自由基清除剂 (N - 2 -mercapto propionylglycine ,MPG)在鼠肝缺血再灌注损伤发病机制中的作用。方法 :Wistar雄性大鼠 2 8只 ,随机分为A、B、C、D四组 :A为空白对照组 ,B为I/R组 ,C为MPG组 ,D为UTI组 ,B、C、D三组分别缺血 6 0min后 ,均再灌注 30min、6 0min、2 4h、72h ,A、B、C、D四组在 30min、6 0min、2 4h、72h检测AST、ALT、LDH以及鼠肝的光镜和电镜切片 ,计算各组生存率。结果 :D组AST、ALT、LDH明显低于B组 (P <0 .0 5 ) ,C组则高于B组 (P<0 .0 5 ) ;光镜和电镜形态学损伤程度由轻到重依次为D、B、C ;术后 3d存活率分别为 :A组 10 0 % ,B组 4 2 .86 % ,C组 0 ,D组 85 .71%。结论 :乌司他丁 (UTI)能有效减轻、改善大鼠肝脏缺血再灌注损伤 ,MPG则加重鼠肝缺血再灌注损伤。 Objective:To investigate the protective effect of Ulinastatin (urinary trypsin inhibitor UTI), and free radical scavenger, N-2-mercaptopropionyl glycine (MPG) on reducing hepatic ischemia-reperfusion injury in rats.Methods:Twenty-eight male Wistar rats were randomly divided into four groups: group A, Nil control group (n=7); group B, ischemia-reperfusion group(n=7); group C, MPG treated group(n=7);group D, UTI treated group(n=7). The latter 3 groups were subjected to 60min of hepatic warm ischemia followed by 30min, 60min, 24h, 72h reperfusion. Serum alanine aminotransferase (AST), aspartate aminotransferase (ALT), lactic dehydrogenase (LDH)level were detected at 30min, 60min, 24h and 72h time points.Liver samples were obtained for light and electronic microscopy study.Results:The AST, ALT, LDH level in group B were significantly higher than in group D but much lower than in group C.The morphological injury changes were mild in group D compared with group C and B. The post ischemia/reperfusion survival rates were improved when treated with UTI.Conclusion:This experiment suggests that UTI in comparison to MPG, significantly ameliorates the hepatic ischemia-reperfusion injury in rats.
