摘要 应用酶活性测定和聚丙烯酰胺浓度梯度凝胶电泳技术 ,研究了 3种不同浓度的重金属胁迫对不同类型水稻萌发种子中α,β-淀粉酶同功酶活性的影响。结果表明 :(1) 0 .0 5 m mol/ L的 Cu2 + ,Cd2 + ,Hg2 + 胁迫使水稻萌发种子中α-淀粉酶活性有所增高 ,而 >0 .0 5 m mol/ L的 Cu2 + ,Cd2 + ,Hg2 + 胁迫则导致水稻各品种萌发种子中β-淀粉酶活性显著降低。 (2 ) Cu2 + ,Cd2 + ,Hg2 + 胁迫使武育粳萌发种子中β-淀粉酶活性随重金属浓度的增高呈先上升后下降的趋势 ,并显著抑制 (尤其是 Hg2 + )扬稻 6号和汕优 818的β-淀粉酶活性。 (3) 0 .0 5 m mol/ L的 Cu2 + ,Cd2 + ,Hg2 +胁迫能明显诱导新的淀粉酶同功酶的产生 ,>0 .0 5 mm ol/ L 的 Cu2 + ,Cd2 + ,Hg2 In this paper,the α,β amylase activity measuring methods and polyacrylamide concentration gradient gel electrophoresis technique were used to study the effect of heavy metal stress on amylase activity in germinating rice seeds.The results showed:(1) 0.05 mmol/L Cu 2+ ,Cd 2+ ,Hg 2+ stress caused α amylase activity in germinating Wuyujing,Yangdao 6,Xianyou 818 seeds to increase obviously.However,the higher concentration of Cu 2+ ,Cd 2+ ,Hg 2+ stress caused α amylase activity to decrease markedly,and the inhibited degree of α amylase activity enhanced with the Cu 2+ ,Cd 2+ ,Hg 2+ concentration decreasing.(2) With the concentration of Cu 2+ ,Cd 2+ ,Hg 2+ increasing,the β amylase activity in germinating Wuyujing seeds increased and then decreased.The activity of β amylase in germinating Yangdao 6 and Xianyou 818 seeds was inhibited by all Cu 2+ ,Cd 2+ ,Hg 2+ treatments (especially,the Hg 2+ ).(3) Although>0.05 mmol/L Cu 2+ ,Cd 2+ ,Hg 2+ stress inhibited the expression of some amylase isozymes which were expressed in normal condition(such as in control),0.05 mmol/L Cu 2+ ,Cd 2+ ,Hg 2+ stress induced the expression of some amylase isozymes significantly.
