摘要
通过分离和出菇试验,获得了纯化的平菇(Pleurotus sapidus)和香菇(Lentinus edodes)单孢系。用溶壁酶去细胞壁制备成原生质体。以PEG为融合诱导剂,诱导两者原生质体融合。1988年获得了可以出菇的融合子。这些融合子形成的子实体,从形态、生长习性和菌伞的颜色特征上都与双亲有明显的差异。大部分氨基酸含量介于双亲之间。同功酶分析也显示出融合子呈现与双亲不同的酶带。融合子出现的上述新性状,可能是平菇与香菇的原生质体基因重组的结果。
Single spore pure lines of homozygous Pleurotus sapidus and Lentinus edodes were isolated and mushroom germination was tested. After removing the cell wall and by using Polye-thyleneglycol(PEG) as fusogen, both the protoplasts were fused to give fusant which could grow up and form mushroom. The fruiting bodies from fusants were remarkably different from that of their parents in morphological characters, growth habits and pileus colours. Amino acid analysis showed that the contents of most amino acids in the fusant were in intermediary between two parents. The isozyme analysis also showed that the fusant manifested itself different zymogram bands from its parents.Based on the comparison with the creteria mentioned above, we believe that the intergrne-ric hybrids have been obtained by manipulating the protoplasms of single spore pure lines of P. sapidus and L. edodes and that the new characteristics of fusants are resulted from the gene recombination of their parent protoplasts.
