摘要
目的:观察实验性糖尿病大鼠海马毛细血管超微结构改变。方法:将成年SD大鼠随机分为正常对照组(5只)、糖尿病组(12只,经腹腔注射链脲佐菌素制备实验性糖尿病大鼠模型)、糖尿病治疗组(12只,给予部分成模大鼠长效胰岛素2-3U/日治疗,使血糖低于10mmol/L),于成模第3月末及第6月末灌注固定取脑,透射电镜下观察海马CA1区毛细血管。结果:糖尿病大鼠海马毛细血管基底膜增厚、内皮细胞与周细胞退变。6月组病变较3月组更显著,各治疗组改变较轻微。结论:糖尿病可导致大鼠海马毛细血管基底膜增厚,早期应用胰岛素治疗可延缓其发生。
Objective: To study the ultrastructure changes of capillaries in hippocampus of diabetic rats and the effect of insulin on these changes. Methods; Sprague-Dawley adult rats(BW 250 g+ ) were divided into normal control group(5 rats), diabetic group (12 rats induced by injection of streptozotocin into peritoneal cavity) and treated group (12 rats, long acting insulin was injected subcutaneously 2-3U per day, fast blood glucose< 10 mmol/L). Rats were killed at the 3rd month and the 6th month. Thin sections were cut with ultrathin microtome and observed under electron microscopes. Results: The basement membrane of capillaries thickened significantly and endothelial cells and pericytes degenerated. These changes on the 6th month group were more significantly than that of 3rd month group. However changes were much milder in the treated group. Conclusion; There are significant pathological changes in hippocampus capillaries of diabetic rats. Early insulin using can delay the occurrence of neuropathological changes of diabetic rats.
