摘要
为探讨粘附分子和炎性细胞因子与SLE发病的关系,用流式细胞术及免疫双荧光染色法,检测了SLE患者淋巴细胞表面CD11a、CD54的表达,同时用放射性免疫法(RIA)检测了血清TNF-α和IL-8的水平。结果表明(1)SLE患者的血淋巴细胞表面CD11a、CD54的表达和血清TNF-α、IL-8的水平较正常对照组显著性升高(P<0.01),且活动期患者高于稳定期患者(P<0.01);(2)SLE伴皮肤血管炎患者淋巴细胞表面CD11a、CD54的表达较SLE不伴皮肤血管炎患者显著性升高(P<0.01);(3)SLE患者血清TNF-α与IL-8的表达呈显著正相关(P<0.05);SLE患者淋巴细胞表面CD11a与CD54的表达呈显著正相关(P<0.05)。外周血淋巴细胞表面CD11a、CD54的表达和血清TNF-α、IL-8的水平均可作为SLE病情活动的参考指标。
To investigate the role of adhesion molecules and cytokines in the pathogenesis of SLE, we examined the expression of peripheral blood lymphocyte adhesion molecule CD11a and CD54 in patients with SLE, using flow cytometric analysis and dual immunofluorescence staining. Meanwhile, the radioimmunoassay(RIA) was used to detect the serum levels of TNF α and IL 8. Results:(1)The expression of CD11a and CD54 on lymphocyte and serum levels of TNF α and IL 8 were markedly increased in patients with SLE (P< 0.01), and all of them were higher in patients with active SLE than those with stable SLE (P< 0.01).(2)The expression of CD11a and CD54 were markedly increased on lymphocytes in SLE patients with cutaneous vasculitis compared with those without the vasculitis(P< 0.01).(3)There were significantly positive correlations between the expression of CD11a and CD54 on lymphocyte in patients with SLE (P< 0.05), and between the levels of TNF α and IL 8 in serum of patients with SLE (P< 0.05).Conclusion: The expression of CD11a and CD54 and serum levels of TNF α and IL 8 are closely related to the activity of SLE and may be used as an index to assess the activity of SLE.
