摘要 目的:优选简便且重现性好的咪喹莫特壳聚糖纳米粒的制备工艺条件。方法:采用壳聚糖交联法制备;用正交试验法对影响制备的三个因素进行优选;用紫外分光光度法测定载药量。结果:制备工艺中采用搅拌时间1h,滴注速度5滴·S^(-1),药物/材料比为1:2,所得咪喹莫特壳聚糖纳米粒载药量最高为5.81%,粒径为543nm,跨距为0.96。结论:优选的制备工艺简便,重现性好,可制得载药量高、形态圆整、分布均匀的咪喹莫特壳聚糖纳米粒。 Objective: To optimize preparation technology, which was simple, reliable, and feasible, and good reproducibility, for imiquimod chitosan nanoparticles. Method: Chitosan-cross linking method was used to prepare imiquimod chitosan nanoparticles. Three factors influencing the quality of imiquimod chitosan nanoparticles were optimized by orthogonal test. The drug,s content was determined by UV-spectrophotometry. Result: When the infusing rate was 5 drops·s-1, mixed duration was 1h, drug-, stuff ratio was 1:2, the highest drug content was achieved . The prepared imiquimod chitosan nanoparticles had an average diameter of 543 nm with a span value of 0.96. Conclusion: The optimal procedures were simple and repeatable with a high drug content. The nanoparticles were round in shape and distributed uniformly.
