摘要
目的 探讨Iodogen法13 1I标记磁共振单克隆抗体靶向对比剂McAb PMP的方法。方法 采用Iodogen法13 1I标记McAb PMP ,并测定标记率、放化纯度、比活度和免疫活性 ,进行动物免疫显像 ;结果 标记率为 89 7% ,放化纯度为 3 16mCi/ml。经RM90 5活度计测定的比活度为 89 6μCi/ μg。在注射13 1I McAb PMP后 ,在结肠癌裸鼠动物模型显像时 ,2 4h出现特异性聚集 ,48h和 168h仍然保持聚集。结论 13 1I McAb PMP在Iodogen碘化标记后能较好地保持其免疫活性 ,特异性聚集在肿瘤部位 ,提示McAb
Objective To investigate the feasibility of employing Iodogen method to label monoclonal antibody targeted contrast McAb PMP with 131 I. Methods After McAb PMP was 131 I labeled with by Iodogen method, labeled rate, purity of radiochemistry, specific activity and immune activation were determined repectively. Radioimmunoimaging was performed on colon cancer of BALB/c nude mice. Results The labeled rate was 89 7%, and the purity of radiochemistry was 3 16 mCi/ml. The specific activity was 89 6 μCi/μg determined with RM905. After the colon cancer mice were injected with 131 I McAb PMP, their emission CT scan appeared peculiar gathering of the contrast in 24 h after injection, which kept even in 48 h and 168 h. Conclusion 131 I McAb PMP keeps its immune activity after Iodogen labeling method, gathered at tumor position in vivo . This result indicates that McAb PMP can be applied to MRI as contrast agent.
