九节龙皂甙对Hela细胞生长的抑制作用 被引量:16

摘要 目的 观察九节龙皂甙 (Ardiusilloside)对人宫颈癌Hela细胞的抑制作用。方法 九节龙皂甙 6 2 5mg·L-1处理Hela细胞 0~ 72h ,在不同时间点采用光镜观察Hela细胞形态 ,以琼脂糖凝胶电泳检测Hela细胞凋亡DNA的梯形带 ;应用特异性Ca2 + 荧光探测剂Fluo 3 /AM在LSCM条件下检测不同时期细胞内Ca2 + 分布情况。结果 光镜下体外培养的Hela细胞在九节龙皂甙作用 2 4h开始出现细胞质皱缩和细胞核凝缩等凋亡的形态学特征。 60、72h均检测出DNA梯形带等凋亡的生物化学特征。诱导Hela细胞48、60、72h内Ca2 + 严重超载且维持在较高浓度水平。结论 九节龙皂甙对体外培养的Hela细胞具有明显的抑制作用且诱导Hela细胞凋亡发生 ;细胞凋亡时细胞内 [Ca2 + Objective To observe the inhibitory effect of Ardipusillide on human cervical carcinoma cells (Hela cells). Methods Ardipusillide at 0.25 mg/L was added to the culture medium of Hela cells and cocultured for 0-72 h. The cells were observed at different time points. Apoptosis was detected with light microscope and DNA electrophoresis, while [Ca 2+ ]i was determined with confocal laser scanning microscopy. Results Changes of early apoptosis were found in Hela cells after treated with Ardipusillide for 24 h, and DNA ladder was found in 60 and 72 h samples [Ca 2+ ]i increased and sustained at arather high level after induced with Ardipusilliae for 60 and 72 h. Conclusion Ardipusillide has an inhibitory effect on Hela cells in vitro and could induce apoptosis. At different time points, intracellular free Ca2+ concentrations are maintained at a sustained increase.
