摘要
目的 提高多价小鼠精子抗原表位多肽的免疫性避孕效应。方法 应用固相化学合成法合成具有两种精子抗原蛋白关键表位的 35肽 ,纯化后的多肽与皂甙QuilA及磷脂、胆固醇混合 ,透析制备多肽 免疫刺激复合物 (ISCOMs) ,并用于免疫雌性C5 7小鼠。结果 合成后的 35肽经纯化后纯度大于 95 % ,经质谱仪鉴定质荷比与理论值一致 ;透析法制备的多肽 ISCOMs在电镜下呈规则的蜂巢样、由多个亚单位构成的开放型结构 ;免疫后小鼠的血清及阴道冲洗液中特异性IgG和IgA最高分别为 1∶180 0 0和 1∶16 0 0 ;同时受孕率降为 2 0 0 % ,显著低于对照组 (P≤ 0 0 1)。结论 多肽 ISCOMs复合物具有独特的蜂巢样结构 ,其应用可显著提升多肽的免疫效应 ,尤其是粘膜免疫 ,获得较好的免疫性避孕效果 。
Objective To evaluate the contraceptive effect of mul ti epitopes peptide of sperm antigens.Methods The multi-epitopes p eptide with 35 amino acid residues was synthesised and purified.After mixed with Saponia A, phospholipid and cholesterol,the polypeptide-immunostimulating comp lexes was established,and the C57 mice were immunized.Results The purity of synthetic peptide was above 95% after purified, and its ratio between quality and charge was equal to the theoretical value.The regulated honeycombs with multi-subunits were observed by electron microscope in the polypeptide-im munestimulating complex.After immunized,the highest level of specific IgG in ser um was 1∶18000,and the IgA in the washing was 1∶1600;and the pregnancy rate de creased to 20.00%.Conclusion ISCOMs could enhance the immune effe cts and raise the efficiency of contraception
