摘要
目的 旨在了解人精子顶体反应率优选及冷冻前后的变化。方法 应用甘油、甘油 卵黄 柠檬酸钠 (GYC)及甘油 卵黄 柠檬酸钠 L 谷氨酰胺 (GYCG)三种冷冻保护剂及上游法、Percoll密度梯度离心法及双曲管优选法 ,检测冻贮前后及优选前后人精子顶体反应率。结果 (1)人精子冻融后其顶体反应率在GYCG组无显著改变 ,而在甘油组、GYC组则明显下降 ;(2 )在上游法、Percoll密度梯度离心法及双曲管优选法优选后顶体反应率均有显著增加。结论 冷冻复温使人类精子顶体反应率下降 ,GY CG对人类精子的顶体反应率有较好的保护作用 ;
Objective In order to understand effect of different cr yoprservation medium and selection techniques on acrosome reacted rate in human spermatozoa.the cryopreservation used in this study were glycerol,glycerol-egg yolk-sodium citrate(GYC) and GYC-glutamine(GYCG).The selection techniques incl ude swim-up, Percoll and double-flection.Observing acrosome reacted rate befor e and after thawing and selectiontechniques.It revealed that:(1)the acrosome rea cted rate decreased no significantly after thawing than before cryopreservation in GYCG group, but decreased significantly in glycerol and GYC;(2)The acrosome r eacted rate increased significantly in swim-up, Percoll and double-flection. Conclusion freezing and thawing decrease the acrosome reacted rat e,GYCG has better prsesrvation effection on human spermatozoa than gl ycerol gro up and GYCD group.The three selections increase significantly the acrosome react ed rate after selection.
