目的 :探讨偏头痛患者血小板 5 HT2A受体mRNA和粘附分子表达的改变及其相互关系。方法 :利用逆转录聚合酶链技术检测偏头痛患者血小板中 5 HT2A受体mRNA的表达 ,应用流式细胞仪检测偏头痛患者全血血小板膜CD41和CD62P的表达 ,并分析 5 HT2A受体mRNA表达与CD41和CD62P的相关性。结果 :发作期组患者 5 HT2A受体的表达水平与间歇期组和正常组有显著性差异 (P <0 .0 1)。发作期组、间歇期组血小板膜粘附分子表达均较正常组高 (P <0 .0 1)。发作期组 5 HT2A受体mRNA与CD41高度正相关 (r =0 .983,P <0 .0 1) ,与CD62P中度正相关 (r=0 .716 ,P <0 .0 1)。结论 :偏头痛患者 5 HT2A受体的表达和血小板膜粘附分子的表达水平均显著增强 。
Objective: To investigate changes in the expression of 5 HT 2A receptor mRNA and platelet membrane adhesion molecules in migraineurs during the periods of attack and intermittence. To study the interrelation between the expression of 5 HT 2A receptor mRNA and platelet membrane adhesion molecules. Methods: The RT PCR method and the image pattern analysis semiquantitation method were used to determine the 5 HT 2A receptor mRNA expression levels in the migraineur groups and the normal control group. Flow cytometry was used to detect the expression of whole blood platelet membrane adhesion molecules in those groups. The interrelation between the expression of 5 HT 2A receptor mRNA and adhesion molecules was analysed. Results: 5 HT 2A receptor mRNA in the attack group was 1.48±0.75, higher than that of the intermittence group (0.41±0.12), and the normal group ( 0.26±0.04 , P<0.01). The expression of whole blood platelet membrane adhesion molecules had a higher increase during the periods of attack and intermittence, compared with the normal control group (P<0.01). The 5 HT 2A receptor mRNA expression level in the migraineur groups had a high positive correlation with CD 41 (r=0.983, P<0.01) and moderate positive correlation with CD 62P (r=0.716, P<0.01). Conclusion: 5 HT 2A receptor mRNA and platelet membrane adhesion molecule expression level during the period of attack was significantly augmented and platelet activation was increased. Platelet activation might act an important role in developing migraine.
