摘要
目的 :观察外用碱性成纤维细胞生长因子 (b FGF)对大鼠深 度烫伤创面愈合过程中增殖细胞核抗原 (PCNA)、p5 3、原癌基因 c m yc和 caspase 3的影响 ,初步认识增殖与凋亡活动对创面愈合的作用。方法 :74只 Wistar大鼠随机分为正常对照、单纯烫伤和 b FGF治疗 3组。利用大鼠 30 %总体表面积深 度烫伤模型 ,于伤后 3、6小时及 1、3、7和 14日采取创面皮肤标本 ,采用免疫组织化学染色技术检测创伤组织真皮内成纤维细胞 PCNA、p5 3、c m yc和 caspase 3蛋白的表达变化情况。结果 :烫伤大鼠创面中成纤维细胞的增殖与凋亡活动与 p5 3、c myc蛋白表达有一定的关系 ;局部外用 b FGF对成纤维细胞的增殖等活动产生影响。结论
Objective:To study the effects of basic fibroblast growth factor(bFGF) on the expression of proliferating cell nuclear antigen(PCNA),p53,cmyc and caspase3 on partial thickness scald injury,and their effects on proliferation and apoptosis of fibroblasts during wound healing.Methods:Seventyfour male Wistar rats were divided randomly into 3 groups as follows:①normal control( n =2);②injuried control group( n =36),and they were further divided into postburn 3 hours,6 hours,1 day,3 days,7 days and 14 days groups;③bFGF group ( n =36):the time course of tissue collection was the same as injuried control group.The immunohistochemistry staining technique was used to detect the expression of PCNA,p53,cmyc and caspase3. Results:The expression proliferation and apoptosis of fibroblasts showed a relationship with the changes in p53 and cmyc.Exogenous bFGF showed effects on proliferation of fibroblasts.Conclusions:Proliferation and apoptosis of fibroblasts are important process during tissue repairing.
