摘要
以人工饲养的北星二号蚯蚓为材料,通过对14种进口和国产离子交换树脂和 DEAE-纤维素的筛选试验和比较研究,确定了蚯蚓纤溶酶的分离纯化的较佳工艺:组织匀浆或磷酸缓冲液保温提取→Amberlite IRA 904或 DEAE-纤维素离子交换柱层析分离→Sephadex G 100或 Sephadtx G 75凝胶层析纯化→真空冷冻干燥得蚯蚓纤溶酶冻干粉。该工艺所需设备简单,操作方便,适合中。
In this study,the authors took artificial fed fresh earthworm (Eisenia foetida)as the experimental material.They have determined the better technology for separation and purification of earthworm fibrinolysin through the choosing experiments and comparative studies of 14 kinds of ion exchnge resins as Amberlite IRC-50,D290,D380 etc, and several kinds DEAE-cellulose.Its main steps are as follows:extract- ing by pounding the fresh earthworm to pieces or incubated with phos- phate buffer solution…separating by ion exchange chromatography with Amberlite IRA-904 or DEAE-cellulose…purifying by Sephadex G-100 or Sephacryl S-300 gel chromatography…freezing dry,getting freeze dried powder of earthworm fibrinolysin.This technology,only requiring simple equipments and manipulating easily,is suitable to produce in a large scale.
