变形链球菌重组质粒pcDNA3-pacA/pcDNA3-pacP免疫定菌鼠的龋齿记分研究 被引量:2

摘要 目的 观察已构建的基因重组质粒 pc DNA3- pac A和 pc DNA3- pac P作为防龋疫苗的防龋效果。方法 分别将基因重组质粒 pc DNA3- pac A、pc DNA 3- pac P和 pc DNA3- pac A+pc DNA3- pac P联合使用作为基因疫苗注入免疫定菌鼠颌下腺区皮下 ,以 Keyes龋齿计分法评估免疫后定菌鼠磨牙的患龋情况。结果 重组质粒单独免疫组和联合免疫组大鼠的龋齿计分均明显低于阴性对照组和空白对照组 (P<0 .0 5 ) ;联合免疫组和 pc DNA3-pac A组的龋齿记分均明显低于 pc DNA3- pac P组 (P<0 .0 5 )。结论 重组基因质粒 pc DNA3- pac A和 pc DNA3- pac P均可有效的控制龋损的发生 ,降低龋坏程度 ,有望成为防龋基因疫苗。 Objective To evaluate the effect of the vaccine of pcDNA3-pacA and pcDNA3-pacP against dental caries. Methods Gnotobiotic rats were vaccinated with pcDNA3-pacA or/and pcDNA3-pacP through submaxillary gland. The keyes caries scores were calculated and compared. Results The caries scores of the vaccinated rats were significantly higher than those of controls (P<0.05). The pcDNA3-pacP produced weaker effect than the pcDNA3-pacA (P<0.05). Conclusion The recombinant plasmid pcDNA3-pacA and pcDNA3-pacP can be used as a novel gene vaccine against dental caries.
