过氧化氢对内皮细胞表面粘附分子表达的影响 被引量:14

摘要 目的 研究 H2 O2 影响内皮细胞与中性粒细胞的粘附机理。方法 采用流式细胞仪检测了 H2 O2(2 5 0 μmol/L)对培养的人脐静脉内皮细胞 (HU VECs)表面粘附分子 ICAM- 1、VCAM- 1、E- selectin表达影响的时程变化 ,和不同浓度 H2 O2 (5 0、15 0、2 5 0、35 0、45 0 μmol/L )作用下内皮细胞粘附分子 ICAM- 1、VCAM- 1、E- selectin表达的变化。结果  1用浓度为 2 5 0 μmol/L 的 H2 O2 处理内皮细胞 ,三种粘附分子表达的时间过程不完全相同 ,I-CAM- 1表达持续上调 ,VCAM- 1表达在 3小时达到峰值 ,而后随着时间的延长逐渐下降 ;E- selectin表达在 3小时才开始显著上调 ,5小时达到峰值 ,然后随着时间的延长逐渐下降 ;2用不同浓度的 H2 O2 处理内皮细胞 3小时后 ,介导三种粘附分子表达达到峰值的浓度不完全相同。ICAM- 1和 VCAM- 1表达在浓度为 2 5 0 μmol/L 时达到峰值 ,而 E- selectin表达的峰值在浓度为 35 0 μmol/L 时 ,当浓度继续增高时 ,其表达明显下调。结论  H2 O2 能引起内皮细胞表面粘附分子表达上调 ,在中性粒细胞对内皮细胞的粘附过程中起着重要作用 ,H2 O2 可能通过激活粘附分子的表达而加重炎症过程。 Objective Cytotoxic products of neutrophil contribute to ischemia reperfusion injury of several tissues, Hydrogen peroxide (H 2O 2), one of the cytotoxic products of neutrophil, also promotes the adhesion of neutrophil to endothelial cells. The objective of the present study was to investigate the effects of H 2O 2 on adhesion molecules expression in cultured human umbilical endothelial cells (HUVEC). Methods Using flow cytometry, we measured the time and concentration dependency of the effects of H 2O 2 on the expression of ICAM-1, VCAM-1, and E-selectin in HUVEC. Results ① The expression processes of the ICAM-1, VCAM-1, and E-selectin varied after treated with 250 μmol/L H 2O 2. While the ICAM-1 expression increased sustainedly, the VCAM-1 expression reached the peak after 3 hours stimulation and then descended gradually, and the E-selectin expression did not increase until 3 hours stimulation and reached the peak at 5 hours. ② The maximal expression of the ICAM-1 and VCAM-1 occurred at the concentration of 250 μmol/L of H 2O 2, whereas it was 350 μmol/L for the E-selectin. Conclusion The promotion of adhesion molecules expression on endothelium induced by H 2O 2 may play an important role in the adhesion between ECs and neutrophils. H 2O 2 may promote the inflammation process by activating the adhesion molecules expression.
