扎那米韦等对流感病毒神经氨酸酶的抑制作用

Inhibition Effects of Zanamivir and Other Five Drugs on Neuramidinase in Influenza Virus

目的 :观察扎那米韦等 6种临床抗流行性感冒或抗病毒上呼吸道感染药物对流感病毒神经氨酸酶(neuraminidase ,NA)的抑制作用。方法 :应用荧光测定法进行体外酶活性测定。结果 :扎那米韦对我国分离的 3株流感病毒粤防 72 2 43、济防 90 15、四川 2 0 0 0 3 8的神经氨酸酶均有抑制作用 ,其半效抑制浓度 (IC50 )分别为 0 .40 ,0 .2 3 ,2 .5 7nmol·L 1 。金刚烷胺、金刚乙胺、利巴韦林 (RBV)、α 干扰素 (α IFN)和中药抗病毒口服液对流感病毒神经氨酸酶无抑制作用。结论

Objective:To observe the inhibition effect of zanamivir and other five drugs on influenza virus neuramidinase (NA) identified in China. Method:Fluorometric assay was used for test the activity of the enzyme in vitro. Results:The IC 50 of zanamivir against NA of influenza virus strains Yuefang 72～243, Jifang 90～15 and Sichuan 2000 38 isolated in this country were 0.40 nmol·L 1 , 0.23 nmol·L 1 and 2.57 nmol·L 1 , respectively. Amantadine, rimantadine, ribavirin, α interferon and Chinese traditional medicine Kangbingdukoufuye did not have any effect on NA of influenza virus. Conclusion:Zanamivir exerted similar inhibition effects on the three strains of influenza virus isolated in China.
