不同固化物对月季试管苗生根影响的研究 被引量:4

摘要 将组织培养苗用于生产,成本太高是个大问题,如何降低成本是当务之急。琼脂做为培养基中的固化物是必不可少的,尤其是用于生根培养基,培养周期一般不超过半个月,利用率较低。本研究从1987年开始进行筛选新的固化物,试图取代琼脂,以达降低成本之目的。 材料与方法 供试月季品种‘火焰’、‘杏花村’、‘丽丽玛琳’的试管苗共418株。固化物有蛭石、珍珠岩、河沙(这3种物质自身携带的矿物质不加考虑)和琼脂。 首先在室内进行筛选做为配制生根培养基中的固化物,并将月季试管苗分别放入筛选出的新的固化物所配制的生根培养基和用琼脂做固化物配制的生根培养基中进行生根率的比较。然后,将分别在两种固化物配制的生根培养基中产生的生根试管苗移栽室外,对其移栽成活率进行比较,并做统计分析。 In this article, the anthor has experimented upon Rosa cvs. in root-striking medium prepared by new supports such as river sand per-lite, and vermiculite respectively with sieve method. The results show that the effectiveness of vermiculite is similar to agar and is better than river sand and perlite when they are used as supports of root-striking media of tissue culture. So, vermiculite has been selected and used for expanded test. In the test, root-striking rate and transplanting survival rate of tube shoots of nine cultivars of rose in vermiculite and agar medium have been tested and compared. No significant differences have been found between the two media in the experimental results have been analyzed by statistical method. In the last part of this article, comparison of the costs between vermiculite and agar has been made. The former is much cheaper than the latter. Therefore, vermi-culite has been chosen as a support of root-striking medium instead of agar for the purpose of reducing the cost of tissue culture and rapid reproduction.
