高效液相色谱法同时测定树鼩脑组织中的去甲肾上腺素,多巴胺,5-羟色胺及色氨酸 被引量:2

Simultaneous Determinations of Norepinephrine Dopamine Tryptophan and 5-Hydroxytryptamine in Tree shrew Brain by High Performance Liquid Chromatography

摘要 应用高效液相色谱(HPLC)--荧光检测,在一根带有预柱的LKB Superpac ODSC_(18)钢柱上,使用离子对系统做为洗脱液,对一组正常活动规律的树鼩脑组织液中的去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)及色氨酸(TRP)进行测定。荧光检测的波长为激发波长280nm,发射波长360nm得到各物质最小检测限量为:40ng NE和80ngDA(信噪比3:1)以及10ng5-HT和15ng TRP(信噪比4:1)。此方法具有实验操作简单、方便、快速、灵敏度较高、样品易于处理等特点。 A high performance liquid chromatography with fluoremetric has been used to determine the concentrations of norepinephrine(NE), dopamine(DA), tryptophan(TRP) and 5-hydroxytryptamine(5-HT) in a group of tree shrew brains with normal active regularity. An ion-pairing system is used as eluant on a LK.B superpac ODS C18(Sμm, 250×4.0mm) stainless steel column connected with a precolumn. The detecting wavelength is excited at 280 nm and emmitted at 360 nm. The detection limits are fond to be 40 ng NE, 80 ng DA(signal to noise 3:1) and 15 ng TRP, 10 ng 5-HT(signal to noise 4:1). The experimental results show that this procedure is simple, convenient, rapid, highly sensitive and it is easy to prepare samples. It provides a reliable method for assaying the quantity of main neurohumores in animal brain and its relation to biological rhythm.
