摘要
为了阐明脾虚证模型大鼠血浆中部分神经肽的变化规律和健脾益气方剂四君子汤对神经肽的调节作用以及对脾虚证病理过程的保护机制 ,应用利血平和苦寒中药方剂小承气汤两种方法复制出脾虚证大鼠模型 ,以特异性放射免疫分析法检测了血浆 β 内啡肽 (β endorphin ,β EP)、降钙素基因相关肽 (Calcitoningenerelatedpeptide,CGRP)和神经肽Y(NeuropeptideY ,NPY)的含量变化。结果表明 :两种方法复制的大鼠脾虚证模型均表现为血浆β EP、CGRP比对照组显著升高 (P <0 0 1) ,NPY比对照组显著降低 (P <0 0 1) ,以上变化经四君子汤治疗后都有不同程度的好转 ,与模型组比较差异极显著 (P <0 0 1) ,接近对照组水平。提示脾虚证的病理变化过程与 β EP、CGRP、NPY等神经肽类的失衡有一定的相关性 ,而健脾益气方剂四君子汤对脾虚证模型大鼠血浆内神经肽含量有较好的调节作用 ,推测这也许是该方能够治疗脾虚证的作用机制之一。
To explore the changes of some neuropeptides and the effect of Sijunzi decoction on neuropeptides in experimantal spleen deficiency syndrome models,the models of spleen deficiency syndrome were established by injected Reserpine and given orally Xiaochengqi decoction respectively.Radioimmunoassay(RIA) was performed to determine the level of neuropeptides.The results indicated that two kinds of model compared with the control group,the contents of β-endorphin (β-EP) and calcitonin gene related peptide(CGRP) incresed respectively in plasma,while the level of neuropeptide Y(NPY) decreased in plasma (all P<0.01).Meanwile after Sijunzi decoction treatment,the contents of β-EP,CGRP and NPY were all recovered to a differnt degree (P<0.01).It is showed that there was obvious imbalance of neuropeptides in rat models of spleen deficiency syndrome and the Sijunzi decoction could regulate the neuropeptides,which might be one of the therapeutical mechanism of treating spleen deficiency syndrome with Sijunzi decoction
